Да. Он работает для всех классов, кроме классов, загруженных bootstrap classloader. Другой способ определить это:

Class klass = String.class;
URL location = klass.getResource('/' + klass.getName().replace('.', '/') + ".class");

Как заметил notnoop, метод klass.getResource() возвращает местоположение самого файла класса. Например:

Метод getProtectionDomain().getCodeSource().getLocation() возвращает местоположение файла jar или CLASSPATH
